Genomics is a new and quickly evolving field with a huge capacity to extend and improve human life. For the most part, DNA carries out its action through proteins. A gene is first transcribed into less stable messenger RNA. Interrupting, or intronic, information is cut out of the messenger RNA and the remaining RNA molecule is sent out of the nucleus and into the endoplasmic reticulum. In the endoplasmic reticulum the messenger RNA is copied again, this time into protein. This final translation is done by transfer molecules, which contain the code for translation, and ribosomes, which line up the messenger RNA and the transfer molecules so that they can interact. The transfer molecules, called tRNAs, are like three-pronged forks. On one end are three letters from the original DNA sequence, a word, written in RNA. The other end holds the corresponding protein monomer, the amino acid. The amino acid that corresponds to each word varies depending on the species. The ribosome lines up the transfer molecule forks with the attached protein monomers along the RNA. The amino acids are conn ected to form a protein, after which the transfer molecules are reused and the messenger RNA is degraded. The cell sends the completed protein product to the Golgi apparatus, the cell’s post office, and the Golgi packages the protein and sends it to its destination inside or outside the cell. The protein then carries out the function prescribed by its encoding DNA, whether it is the keratin in your hair or an antibody in your immune system. Meanwhile the original DNA is safe in the nucleus, in two copies. It never leaves, and it is split apart and replicated only when the entire cell is replicated. The human genome is written in about 3 billion base pairs, or letters. If you stretch out the DNA from one nonreplicating cell, it will be about two meters long (3 billion base pairs in 23 chromosomes · two of each chromosome in the cell · 0.34 nanometers between consecutive base pairs). If you concatenate the DNA from all of your cells and stretch it out as one string, it will reach the sun and back 67 to 333 times, or the moon and back 25,000 to 125,000 times (2 meters of DNA in each cell · about 10 to 50 trillion cells in the human body ÷ 300 million kilometers from the Earth to the sun and back, or 800,000 kilometers from the Earth to the moon and back). In the cell, the DNA is wound tightly around proteins called histones, and for this reason, even though we will try to degrade the proteins, the DNA will precipitate in clumps rather than clean strings when we extract it from a vegetable or fruit. While we watched this video we set up the first steps of the DNA extraction protocol, which contains a convenient 10-minute break. Below is the protocol we used. The students wanted to know what each step does to the DNA, so I’ll try to explain it here as well. Materials: A blender. A mesh strainer with very small holes. If you are alone: A clear cup. It looks really cool if you use a champagne glass. A wooden BBQ skewer or something else with which to stir. One eighth teaspoon table salt. About one cup of cold water. A pinch of meat tenderizer or contact lens solution. (We used meat tenderizer.) Two tablespoons liquid laundry detergent. Use clear laundry detergent. Colored laundry detergent will overpower the color of the fruit or vegetable. About half a cup of something that was once alive. It’s okay if it’s frozen. We tried strawberries, split peas, and corn. The kids were most excited about the strawberries. I thought the peas looked coolest. The frozen corn was not very exciting for anybody. A small jug of rubbing alcohol with at least 95% alcohol content. If you are with 10-20 friends: A bag of small, clear, disposable cups. The more translucent cups are worth the extra money. A bag of wooden BBQ skewers or something else with which to stir. One container of table salt from your kitchen. Gallon jug of cold water, which you brought to school empty and filled with cold tap water in the bathroom before class. Two small shakers of meat tenderizer. You won’t use much of this, but it’s better to have two so that they can both be passed around at the same time. A small bottle of clear liquid laundry detergent. A bag or two or three of something that was once alive, like a fruit or a vegetable. It’s okay if it’s frozen. Several jugs of rubbing alcohol with about 95% alcohol content. You’ll need about as much rubbing alcohol as vegetable or fruit, which might be a lot. (Weird looks at the check-out line come with the vast, yellow and green polka-dotted territory of being awesome.) Leave time to potentially stop by multiple CVSes. Among the materials, rubbing alcohol (isopropyl alcohol) can cause irritation to eyes, skin, or the respiratory system. Isopropyl alcohol vapors can irritate the eyes and the respiratory system, contact with eyes can cause damage and burns, and ingestion or inhalation can cause vomiting, drowsiness, and death. The lethal dose is about one cup. It’s unlikely you’ll be able to drink very much, but if you do you will die. You also don’t want to eat the laundry detergent or get it in your eyes. Procedure: Combine in the blender one part vegetable or fruit, two parts cold water, and the salt. If you’re doing this alone, it’ll be half a cup of vegetable or fruit, one cup water, and one eighth teaspoon salt. If you’re doing this with a group you’ll want to fill the blender and scale up the salt appropriately. Blend on high for 15 to 25 seconds.  It is not important that the water be cold, but it is helpful. Most things, including DNA, tend to be less soluble at lower temperatures. (The exception is proteins, which start denaturing, or losing their structure, at higher temperatures, exposing their hydrophobic parts and forcing them to clump together to avoid surrounding water.) The salt, NaCl, dissolves in the water, separating into the charged ions Na+ and Cl. The Na+ neutralizes the negatively charged DNA, allowing the DNA strands to clump together rather than be repelled by each other’s negative charge. Balance your mesh strainer over a clear cup and pour the liquid contents of the blender through the strainer and into the cup. The cup should be at most half full. If you’re doing this with a group you should divide the contents of the blender equally among the group and line the cups up on the table for the next step. Keep in mind that the goop that comes out of the blender earlier has more DNA in it than the goop that comes out of the blender later. Add two tablespoons of clear liquid laundry detergent to each cup of vegetable goop. If you’re doing this with a group you can use the bottommost line in one of the plastic cups to measure out a very approximate two tablespoons.  The laundry detergent disrupts the membrane enclosing the cell and potentially the nuclear membrane enclosing the DNA. Distribute a vegetable goop cup and a BBQ skewer to each person. Everyone should stir gently and then let the solution stand for 10 minutes. Now is a good time to watch the above 7-minute video. Pass around the meat tenderizer and the rubbing alcohol. Each person should add a pinch of meat tenderizer to their cup and stir gently again, and then add about as much rubbing alcohol as there is vegetable mixture. The rubbing alcohol makes the DNA clump together, since the DNA is less soluble in rubbing alcohol (or any other alcohol) than in water. The meat tenderizer contains protease, an enzyme that degrades the proteins that accompany the DNA. The DNA will appear as white goop on the surface of the green or red goop. You can spin it onto the BBQ skewer like cotton candy, but I think it looks prettier and much less gross if it’s left in the cup.  Here are some photos my students took at the end of the process. The fruits and vegetables used, clockwise from the top left, are strawberries, peas, corn, and mixed berry, all frozen. That no one photographed the DNA extracted from the corn is, I think, additional testament to frozen corn not being very interesting.       Afterwards, I opened the floor to questions and short chalk talks and we ended up going in very interesting directions. I almost wish I’d had an older class so that students could teach each other more than I talked at them, but at the same time it seems like younger people ask more questions and their questions are often more interesting. Some of the things we talked about were transposons, viruses, cancer, stem cells, ribosomes and the RNA world, DNA sequencing technologies, sex chromosomes and their evolution, alternative splice sites, and the evolutionary benefits of aging and death. I got some emails in the following days expressing interest in biology and asking about things we talked about in class, which was such a wonderful feeling. If you have time and a presentation at your local elementary school seems like something you would enjoy, you should ask about trying it. A few weeks ago my mom repeated the presentation and the DNA extraction with my little brother’s fifth grade class, and apparently they asked even more interesting questions. However, some students may continue to struggle in one or more areas, and this can be a matter of concern for parents. The student may also feel discouraged, which could have a further negative effect on his or her performance, plus the teachers may show concern. Dont fear, though. We have four tips to help struggling students perform better at school.    1. Evaluate Time Management Private school can be exciting, especially if the student attends a boarding school. Longer days, more free time, sports and afternoon activities, and more time for socialization. Its important to first and foremost look at the students time management skills. Is he or she devoting enough time to studies, or are other extracurricular activities monopolizing their time? This can often be a quick and simple solution, but helping your child simply create a more regimented schedule that ensures enough time is being spent on studies.   2. Is the Student Studying Right? Going online with time management, students have to develop good study habits in order to succeed in difficult schools. Being bright is not enough. Its important to be inquisitive and to care about what you are learning, but you also must make sure that you are studying efficiently and effectively. That means that you have to use the right tools to help you retain the information, and you must have a well-ordered organizational system that helps you keep track of your work and plan ahead for projects and tests. Many schools offer online learning management systems that can help a student better prepare. Procrastination and cramming do not lead to as positive results as studying over time and planning ahead do. These are good habits to develop for life after school as well. 3. Does the Student Have Learning Issues? Some students struggle because they have undetected learning disabilities that are getting in the way of their performance. Even bright students can have learning challenges, and these issues may be picked up only in the later grades when academic demands on students increase. If parents or teachers believe that a student who has chronic difficulty in school may have a learning issue, the student can undergo an evaluation conducted by a professional. This evaluation, sometimes referred to as an psycho-educational evaluation or a neuropsychological evaluation, helps break down whats getting in a students way in a non-punitive and non-stigmatizing way. Part of the outcome of an evaluation can be recommendations about how a student learns best, including potential accommodations, or changes in a students curriculum, to help him or her. These accommodations can include, for example, extra time on tests, if it is warranted, or use of a calculator on math tests, if allowed. The student must still do the work, but he or she can have support programs in place to help him or her succeed. With these accommodations and help in place, such as the support of a learning specialist or resource room, it may be possible for the student to stay in his or her original school and to succeed. 4. Evaluate the Students Fit with the School While this may be a disappointing solution, sometimes, its the right one. The best private school for any child is  the one that fits him or her the best.  That means that the child can succeed in the school academically, emotionally, and in regards to extracurricular interests. While its not necessary to be the top student, a student should place roughly in the top third or at least half of his or her class, particularly in the upper grades, to have a better shot at college admissions. If the curriculum is far too demanding, the student may not fare as well in college admissions, and, more importantly, the student will not be able to grasp enough of the curriculum to learn the material well and to develop good skills. A student who fits well with his or her school will also be able to develop confidence and a sense of achievement. If a student is not a good fit, he or she may have to  change schools. Some people say that all the good ones are either married or gay. As a single heterosexual woman that has been in the dating scene, I must say that statement is not far from the truth. I had been a mother for two years when he was tossing his high school graduation cap in the air. Needless to say, I politely ended his visit with no goodnight kiss included. Another recent dating encounter was the dreaded blind date. As I drove to the agreed upon meeting place all I could think was â€Å"Why do I get myself into these situations? † Yet, the guy I met was really nice. He was extremely attractive and had a great personality. I was pleasantly surprised at how easy the conversation flowed from subject to subject without any awkwardness that I had previously experienced. As the gathering was breaking up, he asked if he could have my telephone number and when would be a good time to call. Of course, I gave him my number and tried not sound eager as I said, â€Å"Anytime would be fine. † After he left, I overheard a conversation regarding his astonishingly recent separation from his wife. He was married. My newly elated attitude towards dating was quickly deflated. Transitioning from the â€Å"in a relationship† world into the â€Å"unattached† world is not as transparent as I once thought. I have learned that men in my dating pool tend to be scorned by women from their previous relationships. Those scorned men are almost always cautious to the point of being noncommittal. I have never enjoyed cleaning especially a mess created by someone else. When it comes to love, I just don’t want to work quite that hard. After all, love is supposed to be easy in the beginning, isn’t it? Being single is hard but being with the wrong man is even harder. The negative aspect of alcohol and drug use in television is that it sends a negative message to teenagers, its says its cool to drink and smoke. According to Kids Health.com kids and teens ages eight to eighteen spend nearly four hours a day watching television ( The Nemours Foundation, 2010). This is about three times the time amount they spend listening to the radio. Children are greatly influenced by what they watch and many of them idolize stars from these shows and want the mimic what they watch, especially if the use of alcohol, nicotine and illicit drugs are glamorized. These shows depict unsafe behavior and often there are no consideration about the consequence of on the use of alcohol, smoking and use of drugs. Alcohol and nicotine seem to be a favorite among prime time shows; illicit drug use is mostly portrayed in movies. Illicit drugs use and alcohol are hyped up in movies more so than in prime time, but some movies also send a message how drugs can lead to destruction and the struggle to overcome addiction. In 1995, The Basketball Diaries staring Leonard DiCaprio portrayed the true story of Jim Carroll a teenagers struggle with heroin addiction whose only desires are the game of basketball and heroin. In The Basketball Diaries Jims the only escape from a best friend that is dying and a coach who is a child predator is heroin. In a downward spiral Jims addiction for heroin leads him into the streets where he steals, robs and prostitutes himself. It is only through a friend and his relentless pursuit of his desire for Jim to be clean that he overcomes his addiction and fulfills his dreams. The message in The Basketball Diaries relays how drug addiction can destroy your dreams lead you int o a path of crime and self destruction. The message in this movie was a detail of how a lifestyle can easily be misled through the use of drugs and the struggle a person will have to take to overcome that addiction. This move portrayed a very effective message; drugs can destroy all your dreams, lead you into a life where basically you would sell your soul to get your next hit. It also showed the struggles to overcome addiction and how positive your life can become once you decide to make positive choices in your life. As much as alcohol and drug use may be glorified in prime time television, there are also antidrug advertisements that try to educate parents and children on peer pressure and the effects of drug use. In 1998 Congress created The National Youth Anti-Drug Media Campaign in collaboration with Partnership for a Drug-Free America the goal of this campaign is preventing and reducing youth drug abuse across the nation. The main goal of this campaign is to create advertisements that deliver antidrug use and the dangers of that behavior to youths. According to The Partnership Attitude Tracking Study (PATS) 2009 there is a correlation between increased teen exposure to anti-drug messages on television and a decreased likelihood of trying drugs over the past ten years. Four out of ten teens agreed that anti-drug messages made them more aware of the risks of using drugs and less likely to try the drugs (Drug Enforcement Administration, 2010). In one of the many advertisement the campaign has pr oduce, The Human Puppet the advertisement depicts a girl is passed out on a chair with her friends gathered around her, they are painting her face and have strapped her arms with scarves and are moving her hands as she is a puppet. The girl is in an unconscious state and never reacts to what her friends are doing to her they make reference to the fact that she is passed out and thrashed and that she will have a major headache in the morning. Eventually they walk away from her, leaving her slumped in the chair (The National Youth Anti-Drug Media Campaign, 2009).This was an excellent advertisement, and depicted the reality of what can happen when you consume alcohol as a teenager and the embarrassing situations that you place yourself in. The advertisement also pointed out the reality of what your friends can and will do to you when you passed out from alcohol use. The one weakness that was noted in this advertisement was that some kids may think that it was more humorous than educati onal. There should have been more depiction of what can happen when a kid drinks too much alcohol such as loss of body function or alcohol related illnesses. Overall these campaigns have been shown to work The National Youth Anti-Drug Media Campaign began in July 1998 and its primary focus was on reducing marijuana use among adolescents. Introduction The collective space vision of all the world’s countries at the moment seems to be Mars, Moon, space colonization, permanent settlement etc. Even Steven Hawking does not think that humans will survive another 1000 years ‘without escaping beyond our fragile planet’. Architects are designing colonial habitats on Mars and Moon creating large flimsy structures for permanent settlements. But is that space architecture is all about? Just to create a new living environment for rats abandoning a sinking ship? Obviously space architecture can be something more than just a permanent habitat in uninhabitable environment.
